[Principle]

This reagent is prepared based on the colloidal gold immunochromatographic test (GICA) principle to detect the content of pregnancy-associated glycoproteins (PAGs) in bovine serum samples. After adding to the sample well, the sample moves along the chromatographic membrane together with the colloidal gold label. If pregnancy-associated glycoproteins (PAGs) are present in the sample, they bind to the colloidal gold marker and antibodies on the detection line, resulting in a purplish-red color. If the PAGs content in the sample falls below the detection threshold, no color reaction occurs.

[Storage and Shelf Life] Store in a cool, dry place (2-30°C), shelf life 24 months.

Serum: Draw 2-3ml of blood as usual and place it in a clean, dry test tube, let it stand for about 1 hour until coagulated, then centrifuge at 4000 rpm for 10 minutes (blood can also be left to stand for about 2 hours until coagulation and serum naturally precipitates), then separate the serum. The serum must be clear, without hemolysis or contamination. Serum samples can be stored at 2~8°C in the short term, and at -20°C in the long term.

[Test Method]

1. Serum sample loading method: Tear open the aluminum foil packaging bag of the test card, remove the test card, place it on a flat, clean table, and use the pipette inside the bag to slowly add 2 drops of serum into the sample loading well.

2. Place at room temperature for 10-15 minutes to determine the result; color development after 15 minutes is ineffective.

[Result Judgment]

1. Negative: In the observation hole, only a purplish-red line appears in the control line area (C).

2. Positive: In the observation hole, both the detection line (T) and the control line area (C) show purple-red lines.

3. Ineffective: In the observation hole, no red line appears in the reference line area (C).

[Interpretation of Test Results]

Pregnancy-associated glycoproteins (PAGs) are a marker for conception in ruminants, and PAG concentrations exhibit characteristic changes depending on the reproductive physiological stage of the cow. The average estrus cycle for dairy cows is 21 days. At the beginning of the estrus cycle, PAGs are at a low level. After successful conception, their serum levels gradually increase. Starting from day 28 after successful conception, the serum PAGs content in non-pregnant cows remains at a low level, while the PAGs content in pregnant cows is at a detectable high level. This product is designed and developed based on these patterns, with testing conducted on day 28 after breeding. If the test result is negative, a retest can be conducted after one week. If the result is still negative, it can be confirmed that the cow is not yet pregnant, and it is recommended to promptly conduct secondary breeding; A positive test indicates that the cow being tested is pregnant and should enter gestation management.

[Limitations of the Test Method]

This test card is for initial screening only and cannot be used as a confirmation method.

2. Do not use other animal serum or other liquids that are not required for sample addition.

3. Products that are expired or have damaged aluminum foil bags must not be used.

4. When removing the test card from the refrigerator, it should be returned to room temperature before opening. Once opened, the test card should be used as soon as possible to avoid deactivation after moisture.

5. Avoid using contaminated, cloudy, severely hemolytic, or large blood lipid samples.

6. Avoid contact with the chromatography film at the sample holes and detection windows on the test card.

7. Waste from the test should be considered pollutants and properly handled according to local regulations.
